We described an aspartyl thioester unnatural amino acid and its incorporation into a recombinant protein. Its utility was demonstrated in two publications by native chemical ligation. Attempts to transform the aspartyl thioester into an asparagine-linked glycoprotein with a natural linkage were unsuccessful.
