SUMMARY:PChem Seminar
LOCATION:Fulmer Hall
DESCRIPTION:PChem Seminar, Friday, 9/17/21 at 4:10 in Fulmer 150\n\nSpeaker:  Michael Martinez\n\nGroup:  Brozik\n\nTitle:   Thermodynamics of Redox Dependent CPR Insertion into Biomimetic Endoplasmic Reticulum Membranes\n\nAbstract:\n\nRedox state dependent confirmational changes of cytochrome P450 reductase (CPR) has previously been shown to change the diffusion of fluorescently labeled single proteins through a biomimetic of the endoplasmic reticulum. The diffusion of the oxidized form changes from 2.5 to 1.5 µm2 s-1 upon reduction via NADPH.  During these experiments the limitations of single particle tracking (SPT) and total internal reflectance fluorescence (TIRF) microscopy showed a difference in the CPR solution-lipid equilibrium between the reduced and oxidized forms, but no way to measure it.  This study focuses on pursuing that difference via fluorescence cross correlation spectroscopy and single molecule tracking to measure the concentration within solution and membrane phases.  By measuring the temperature dependence the standard states Gibbs Free Energy, Enthalpy and Entropy for CPR partitioning into the membrane was also measured.\n\n**There will be no Zoom component to this seminar
