Title: Thiocarboxylates in the Thiol Redox Proteome

Abstract: Thiol redox proteome involves a series of post-translational modifications of proteins, mainly in cysteine. These modifications are known to regulate protein function and alter the thiol oxidative and reductive states of cysteine residues.  A new focus has shifted to thiocarboxylates as an effective sulfur-containing substitute to cysteine for such post-translational modifications at the c-terminal. In this review, we discuss the various methods of thioacid synthesis, the amide bond reactions with these thioacid groups, and the assembly of proteins and peptides. Additionally, applications of these modifications are explored in the biological proteome; a 2010 study suggests that these modifications are useful for the detection of protein thiocarboxylates in bacteria.
