Title: Stochastic Biophysics of More Natively Folded Recombinant Human Aquaporin-4 Orthogonal Array of Particles: A Protein Tracking Study
Abstract: Aquaporin-4 (AQP4) is the central nervous system (CNS) water channel protein in charge of controlling water homeostasis and neuronal activity. AQP4 exists in two main isoforms in the CNS, M1 and M23, with M23 being the isoform that favors stabilization by aggregation into orthogonal arrays of particles (OAPs). What role these OAPs play in the membrane of cells is still largely unknown and the dynamics of OAP assembly in the plasma membrane remain elusive. Through a collaboration with the Nicchia group from the University of Bari, techniques have been developed using insect cell lines to isolate and express the M23-AQP4 isoform in its authentically folded state in an in vitro environment. The goal of this research is to take natively folded AQP4 aggregates, and using both in vitro and in vivo t-+echniques, discover how different environmental cues effect OAP assembly kinetics and equilibrium states.
